ConsiderableNanoarchaeotanoveltyanddiversitywereencounteredinYellowstoneLake,YellowstoneNationalPark(YNP),wheresamplingtargetedlakefloorhydrothermalventfluids,streamersandsedimentsassociatedwiththesevents,andinplanktonicphoticzonesinthreedifferentregionsofthelake.Significanthomonucleotiderepeats(HR)wereobservedinpyrosequencereadsandinnearfull-lengthSangersequences,averaging112HRper1349bpcloneandcouldconfounddiversityestimatesderivedfrompyrosequencing,resultinginfalsenucleotideinsertionsordeletions(indels).However,SangersequencingoftwodifferentsetsofPCRclones(110bp,1349bp)demonstratedthatatleastsomeoftheseindelsarereal.ThemajorityoftheNanoarchaeotaPCRampliconswereventassociated;however,curiously,onerelativelysmallNanoarchaeotaOTU(71pyrosequencingreads)wasonlyfoundinphoticzonewatersamplesobtainedfromaregionofthelakefurthestremovedfromthehydrothermalregionsofthelake.ExtensivepyrosequencingfailedtodemonstratethepresenceofanIgnicoccuslineageinthislake,suggestingtheNanoarchaeotainthisenvironmentareassociatedwithnovelArchaeahosts.Definedphylogroupsbasedonnearfull-lengthPCRclonesdocumentthesignificantNanoarchaeota16SrRNAgenediversityinthislakeandfirmlyestablishaterrestrialcladedistinctfromthemarineNanoarcheotaaswellasfromothergeographicallocations.
