We designed species-specific primers for bog turtles (Glyptemys muhlenbergii) and Burmese pythons (Python molurus bivitattus) and sequenced the amplicons to ensure the primers only amplified the target species. We also provide recommendations for future eDNA applications such as annealing temperature, the number of amplification cycles, and the prevention of DNA contamination
