Exploration of the Imperiled Biodiversity of Mammoth Cave KY Using Environmental DNA. Research Permit and Reporting System
Water samples will be filtered from at least 20 points within Mammoth Cave and from surface springs. Water samples will be filtered on a 0.7µm glass fiber filter and preserved in the field at 20 sites. Environmental DNA will be extracted and amplification of a small portion of the cytochrome c oxidase I (COI) gene will be performed using published primers. Amplicons will be sequenced and a species list will be generated from each sample indicating what species were detected at each locality.
