Field Methods: We will walk on established trails to all three sites. Algal samples will be small (<20 grams each) and the sites of removal of the thin film of cyanobacteria will be inconspicuous. Samples will be placed in sterile bottles. Upon returning to the laboratory, all material will be examined microscopically and the strains will be characterized and photographed. Semi-permanent slides will be prepared (Transeau's syrup medium). Material will also be preserved in both alcohol and formaldehyde. Material will be spread on Z-8 agar-solidified medium to attempt culture. Material will also be immediately prepped for single-filament/colony PCR. Multiple reactions will be needed to obtain multiple loci.
