eDNA samples were taken from three vernal pools in the Cowles Bog, INDU on 5/22/2020 and 6/24/2020. Between 11-15 samples per pool per time point were taken, each consisting of 250mL of surface water filtered through a 0.8 micron cellulose nitrate filter. Due to high sediment load in the water, many samples require several filters. Samples were extracted using a modified Qiagen DNeasy protocol and are being stored in a -20 freezer for qPCR and metabarcoding in spring 2021.
