This study examined six populations of Ptomaphagus hirtus from the Mammoth Cave area in order to obtain insight into the evolutionary relationships among the various populations and to estimate the genetic variations in populations. Individuals were collected from Detrex, Horseshoe, and Danger Caves near Bowling Green, KY; from Wisdon and Copelin Caves near Mammoth Cave National Park; and from White, Running Branch, and Great Onyx Caves in Mammoth Cave National Park. Methodology is detailed. Results are explained. An evolutionary pattern is determined and genetic variations are discussed. Allozymic variation in 12 proteins encoded by 13 loci was analyzed in approximately 900 individuals of the troglobitic beetle Ptomaphagus hirtus, representing six geographically isolated populations in south central Kentucky. Identification of alleles was confirmed by the analysis of about 300 intra- and interpopulation hybrids.
